E-SURE : Hemoglobin E Screening Test
1.Use and property
E-sure is a qualitative screening test for Hemoglobin E which is an abnormal beta globin chain of hemoglobin of red blood cell. By using unique buffer solutions and with the electrical charges binding property, only Hemoglobin E is eluded from the column and has color reaction that can be seen in test tube.
2.Brief information
Hemoglobin E, one of beta thalassemia, has a prevalence of 10-50%. When a Hemoglobin E carrier(heterozygous) or homozygous Hemoglobin E(EE) is married to a Beta thalassemia trait, the child may be born with Hemoglobin E –beta thalassemia disease. Hemoglobin E –beta thalassemia disease is a chronic disease that can have many complications and may not be cured.
Screening Hemoglobin E in couples helps doctors in genetic counseling especially in case of Hemoglobin E –beta thalassemia disease, termination of pregnancy may be considered.
An effective screening and diagnostic test is required to the successful prevention of thalassemia in the population.
3.Principle
Ionic exchange is the principle of this test. When hemoglobin is in specific solution, it has electrical charges. When the solution is passed through column, all hemoglobin are caught by the column. Then a second solution is passed through anion exchanger column, only Hemoglobin E is eluded from the column and can be visualized as red color in test tube. The color intensity depends on the concentration of Hemoglobin E.

5. Preparation
1. EDTA venous whole blood sample (can be kept 2-8C for 3days)
2. whole blood sample is not recommended to be frozen
6. Notice
1. this product can be kept 1-30C
2. this product should be kept away from heat and moisture
3.Expired column and solution should not be used
7. Caution

8.Instructions
1.Open cap of microcolumn, pour buffer in the column out, and disconnect end plug
2.Place microcolumn in test tube that provides in package
3.Pour buffer A 2ml (push 1 time) into microcolumn
4. Pipette whole blood 10 uL. into microcolumn
5. Wait until buffer A totally pass through microcolumn about 10 minutes
6. Pour buffer B 4ml (push 2 times) into microcolumn about 40 minutes
8. Wait until buffer B totally pass through microcolumn
9.Interpretation/analysis

Washing for Re-use
Wash1 : Place used microcolumn in test tube that provides in package
Pour Washing1 solution 2 ml (push 1 time) into microcolumn and wait until Washing 1 totally pass through microcolumn
Wash2 : Pour Washing2 solution 6 ml (push 3 times) into microcolumn and wait until Washing 2 totally pass through microcolumn
Pour Washing2 solution 2ml (push 1 time) into microcolumn, For preserve microcolumn before use again next time.
* microcolumn can be reused up to 10 times*
9. Interpretation/analysis
Homozygous HbE (EE) : strong red color in test tube
Homozygous HbE (EE) : light red-pink color in test tube
10.Limitation
1.This product is a screening test, a diagnostic or confirmation test should be considered by physician.
